| 演 題 : | Studies of nuclear DNA replication fidelity |
講演要旨
Coordinated replication of the two DNA strands of the eukaryotic nuclear genome is catalyzed by DNA polymerases a, d and e. These replicases are related, yet they differ in protein partnerships and physical and biochemical properties, including fidelity. This talk will focus on studies of replication fidelity in budding yeast using replicase variants with error signatures that allow the functions of Pols a, d and e in leading and lagging strand replication to be deduced. Emphasis will be on the rates at which errors are generated during replication of undamaged DNA, and the efficiency with which they are corrected by DNA mismatch repair. Despite wide variations in both processes, the replication and mismatch repair machineries operate in a complementary fashion that ultimately achieves highly accurate replication of both DNA strands of the yeast nuclear genome.
